L’Institut Pasteur de Lille abandonne sous sa forme actuelle l’essai clinique qu’il tentait de mener pour mesurer l’efficacité d’un médicament, le clofoctol, contre le Covid-19, faute de patients volontaires, a expliqué jeudi 9 décembre son directeur général à l’AFP, confirmant une information de la Voix du Nord. «Nous avons mangé notre budget et inclus trop peu de patients, nous allions dans le mur», a indiqué le professeur Xavier Nassif, disant «travailler sur un changement de stratégie pour que cet essai soit repris».
«Nous travaillons sur la façon dont on pourrait répondre à la question “Est-ce que le clofoctol est un inhibiteur de la réplication du Sars ?” en changeant de stratégie, en trouvant un partenaire qui pourrait le faire dans d’autres circonstances, peut-être chez des malades hospitalisés ou à l’étranger», a ajouté Xavier Nassif.
L’Institut avait annoncé en septembre rechercher «entre 350 et 700 patients», âgés de plus de 50 ans, non-vaccinés et présentant au moins un symptôme du Covid-19, pour cet essai clinique. Mais trois mois plus tard, il n’en a recruté qu’une douzaine, malgré l’ouverture de centres dans les Hauts-de-France et aux Antilles. «Trop peu pour qu’on puisse arriver à un début de conclusion», déplore le professeur Nassif. «Le Français ne sont pas très volontaires pour participer à des essais cliniques et les gens de plus de 50 ans et qui aujourd’hui ne sont pas vaccinés ont un petit côté rebelle».
Cinq millions d’euros financés par LVMH
Labellisé «Priorité Nationale de Recherche», cet essai visait à «évaluer la sécurité d’emploi, la tolérance et l’efficacité du clofoctol (…) chez des patients atteints de la Covid-19 symptomatique au stade précoce», selon un communiqué alors diffusé par l’Institut. Le médicament devait être administré aux patients sous forme de suppositoire, à raison de deux par jour pendant cinq jours. Le clofoctol a été commercialisé en France et dans d’autres pays européens, dans une autre indication que celle du Covid-19.
L’Institut avait reçu cinq millions d’euros du géant du luxe LVMH pour financer cet essai. Il mène par ailleurs un autre projet de recherche pour trouver un traitement anti-protéase, qui agirait sur une enzyme spécifique du Covid-19. Ce projet «avance bien» mais comme il porte sur un médicament qui n’existe pas encore, «il va demander du temps, plus que pour repositionner un médicament» comme le clofoctol, souligne Xavier Nassif. L’Institut travaille également à un vaccin administré par voie nasale.
